Assessment of PI3K/mTOR/AKT Pathway Elements to Serve as Biomarkers and Therapeutic Targets in Penile Cancer

چکیده
The PI3K/mTOR/AKT pathway might represent an intriguing option for treatment of penile cancer (PeCa). We aimed to assess whether members this serve as biomarkers and targets systemic therapy. Tissue primary from treatment-naïve PeCa patients was used tissue microarray analysis. Immunohistochemical staining performed with antibodies against AKT, pAKT, mTOR, pmTOR, pS6, pPRAS, p4EBP1, S6K1 pp70S6K. Protein expression correlated clinicopathological characteristics well overall survival (OS), disease-specific (DSS), recurrence-free (RFS) metastasis-free (MFS). AKT inhibition tested in two primarily established, cell lines by capivasertib analysis viability chemotaxis. A total 76 surgically treated invasive were included. Higher significantly more prevalent high-grade tumors predictive DSS OS the Kaplan–Meier analysis, independent predictor worse multivariate regression Treatment pan-AKT inhibitor induced a significant downregulation both pAKT decreased Selected protein candidates mTOR/AKT signaling demonstrate association histological parameters patients, whereas appears be most promising one.

MiRNAs as biomarkers and therapeutic targets in cancer.
The knowledge that miRNA expression is frequently dysregulated in cancer has uncovered an entirely new repertoire of molecular factors upstream of gene expression, with exciting potential as novel biomarkers and therapeutic targets in cancer. Exploiting the unique characteristics of these molecules including their stability, tissue specificity, ease of detection and manipulation, will bring cli...
متن کاملMicroRNAs as predictive biomarkers and therapeutic targets in prostate cancer.
Prostatectomy or irradiation is the most common traditional treatments for localized prostate cancer. In the event of recurrence and/or metastasis, androgen ablation therapy has been the mainstay treatment for many years. Although initially effective, the cancer inevitably recurs as androgen-independent PCa, a disease with limited effective treatments. Enhanced predictive biomarkers are needed ...
متن کاملExosomes as Biomarkers and Therapeutic Targets in Pancreatic Cancer
Pancreatic cancer (PC) is a highly malignant tumor with a poor prognosis; indeed, in recent years the incidence and mortality rate have increased. However, the lack of highly sensitive and specific molecular diagnostic markers makes it difficult to classify PC. Exosomes, cell-derived vesicles that contain bioactive molecules, play key roles in cell−cell communication, a process that regulates d...
متن کاملExosomal miRNAs as cancer biomarkers and therapeutic targets
Intercommunication between cancer cells and with their surrounding and distant environments is key to the survival, progression and metastasis of the tumour. Exosomes play a role in this communication process. MicroRNA (miRNA) expression is frequently dysregulated in tumour cells and can be reflected by distinct exosomal miRNA (ex-miRNA) profiles isolated from the bodily fluids of cancer patien...
متن کاملCancer stem cells: therapeutic targets
Cancer stem cells (CSCs) have been identified as rare cellular populations in many cancers, including leukemia and solid tumors. This minor subpopulation of cancerous cells is immortal tumor-initiating cells which thought to be responsible for cancer initiation, progression, metastasis, recurrence and drug/radiation resistance. Low proliferative rate, high self-renewing capacity, differentiatio...
